1 · Why this matters
Speculative If the handedness of life was determined rather than drawn — if some asymmetry in the physics fixed L-amino acids and D-sugars before selection had anything to act on — then a single number decides it, and that number is measurable. The conditional comes first because it is the honest order: no experiment has measured the parity-violating energy difference between two enantiomers, and section 7 says what would. If instead the sign was drawn, the reason life is left-handed is that a coin landed, and the interesting quantity is not why L but how a per-cent-level bias became an exclusive one.
Established The magnitude is best stated as the ladder of enantiomeric excesses anyone has actually measured, because the gap between its rungs is the whole problem. Circularly polarised light acting on racemic isovaline films at 192 nm produces up to about 2% enantiomeric excess, against a theoretical ceiling of about 1.6% set by the molecule’s own anisotropy spectrum. Aqueous alteration on a meteorite parent body reaches about 10 to 18% in L-isovaline. Crystallisation of an RNA precursor on a magnetised magnetite surface reaches about 60%, and 100% after a second crystallisation. The Soai reaction climbs from 0.00005% to above 99.5% in three consecutive rounds. Biology sits at effectively 100%, in every organism, for something like four billion years. Frontier The prize is a route from the first rung to the last that runs in one vessel on prebiotically available materials with nobody lifting the flask between stages, and no such route has been published.
Frontier Who collects is not the chemical industry, which solved its own version of this problem without needing an origin story. The people who need the answer are the ones deciding what instruments fly: enantiomeric excess is the most-cited agnostic biosignature, and its meaning depends entirely on which answer is right. Speculative If the sign is determined by physics, a mirror-image biosphere is impossible and the sign of an excess says nothing about whether a sample represents a second origin of life. If it is contingent, it is a fair coin, and a D-amino-acid biosphere on an ocean moon would be close to proof of independent genesis. One laboratory measurement changes what a flight instrument’s output means.
Handwave The condition, stated plainly: this upside is real only if one mechanism is responsible end to end, and the field currently has five that each work in a laboratory and none that has been shown to hand off to the next. Section 5 is about why the hand-offs are the expensive part, and section 15 is about the claims that skip them.
2 · Concept overview
Homochirality is the observation that life uses one mirror form of its chiral building blocks and not the other. All nineteen chiral amino acids used in ribosomal protein synthesis are L; the ribose in RNA and the deoxyribose in DNA are D. These are not enrichments. To the limit of measurement they are exclusive, in every organism ever examined, and the exclusivity is the thing that needs explaining.
Established The problem decomposes into three questions that are routinely run together and should not be. First, symmetry breaking: what produced a non-zero enantiomeric excess in a pool that chemistry says should be racemic. Second, amplification: what took a small excess to effective purity. Third, transfer and maintenance: what propagated one molecular class’s handedness into the others, and held it against thermal racemisation over geological time. Frontier Each of the three has candidate mechanisms that demonstrably work in a laboratory. No published sequence runs all three on the same molecules, in the same vessel, without a chemist intervening between stages.
Established Five families of driver are on the table, and they divide on one question: is the sign of the excess predictable in advance? Parity violation in the weak interaction is the only mechanism that makes the same enantiomer favoured everywhere in the universe, because the weak force is the only known interaction that distinguishes left from right absolutely. Frontier Circularly polarised light from scattering off aligned dust in star-forming regions sets a sign locally, and magnetised mineral surfaces acting through the chiral-induced spin selectivity effect set a sign from the direction of the magnetisation — both predict a bias but not which bias. Speculative Crystallisation phenomena — eutectic enrichment, attrition-driven deracemisation, conglomerate formation — amplify a sign without choosing it, and pure statistical fluctuation plus autocatalysis, the mechanism Frank wrote down in 1953, chooses nothing and amplifies whatever fluctuation is largest.
Frontier That division is more useful than the usual taxonomy, because it is the one an experiment can settle. A deterministic mechanism predicts that any independently originated biosphere built on the same chemistry has the same handedness. A contingent one predicts a coin flip. Speculative The two hypotheses are distinguishable by a single laboratory measurement on Earth — the parity-violating energy difference — and, much more slowly, by finding a second biosphere and reading its sign.
Frontier The framing this page tests is that homochirality is an amplification problem rather than a symmetry-breaking problem, and it survives with one serious qualification. Small excesses are easy: several mechanisms make them, and one of them has been measured directly in the laboratory at 2%. Speculative The qualification is that every demonstrated route from a small excess to an exclusive one runs on a substrate that is either not prebiotic, not the molecule life uses, or not connected to anything downstream — and the field has no agreed statement of what an abiotic system is not allowed to do, which means it also has no negative control.
3 · Current scientific position
Established Start with the anchor this brief was commissioned on, because checking it produced two findings. The commission describes “Murchison isovaline enantiomeric excess up to about 15%; the Soai reaction amplifying 0.00005% to above 99.5% in one autocatalytic run”. The second clause is wrong as written, and the first is imprecise in a way that matters.
Established The Soai amplification from 0.00005% enantiomeric excess to above 99.5% required three consecutive rounds of asymmetric autocatalysis, not one. The 2003 result of Sato, Urabe, Ishiguro, Shibata and Soai is described by the reaction’s own discoverers, in their 2022 review and again in their 2022 Royal Society of Chemistry volume, as amplification “by three consecutive cycles” from an initial excess of about 0.00005% — roughly five excess molecules in ten million. Established The single-round amplification factor is far more modest and was measured first: the original 1995 experiment took 2% to 10% in one cycle, and reached 88% only after four. Frontier The correction is not pedantry. A mechanism that requires a batch to be isolated and resubmitted three times is a different prebiotic proposition from one that runs to completion once, and the difference is exactly the hand-off problem section 5 is about.
Established On isovaline, about 15% is neither the largest value in the literature nor the best current measurement of Murchison. The commission’s number sits at the top of the 10 to 15% range reported for the 2021 Aguas Zarcas and Murchison study and understates the 2009 result it is presumably drawn from. Established Glavin and Dworkin reported L-isovaline enrichments in aqueously altered CI and CM chondrites that the 2021 Lunar and Planetary Science abstract, by the same group, quotes as “up to about 18%” for Murchison. Established The 2023 asymmetric-photochemistry paper puts the range across carbonaceous chondrites at “up to about 20%”. Established Meanwhile the best modern measurement of the meteorite the anchor names gives L-isovaline at 10 ± 1% in Murchison and 11 ± 6% in Aguas Zarcas, by ultra-high-performance liquid chromatography with fluorescence detection and quadrupole time-of-flight mass spectrometry. Frontier So the honest statement is a range bounded by method and sample — about 10% measured, about 18 to 20% at the literature maximum — rather than a single figure, and a brief that quotes one number for this quantity is quoting a choice.
Established The third problem with the anchor is the molecule. Isovaline is an alpha-methyl amino acid. It has no alpha hydrogen, which is why it resists racemisation and why its excess survives to be measured; it is also not used by biology. Established The proteinogenic amino acids in the same meteorites carry much larger apparent excesses — L-glutamic acid at 48 ± 2% in Murchison and 73 ± 1% in Aguas Zarcas, L-alanine at 37 ± 4% in Aguas Zarcas — and those are the values nobody treats as extraterrestrial, because terrestrial contamination produces exactly that pattern. Frontier The field’s strongest chirality evidence therefore comes from the molecules life does not use, and its largest numbers come from the molecules it does and cannot be trusted.
Established The excess tracks aqueous alteration, which relocates the mechanism from space to the parent body. That is the explicit finding of the 2009 paper, whose title is about enrichment by aqueous alteration rather than about delivery. Established The 2023 measurement of isovaline’s circular dichroism closed the loop from the other side: pristine, weakly altered meteorites such as Paris and Winchcombe are “racemic or nearly racemic”, while heavily altered ones are enriched. Frontier The delivered-excess hypothesis in its simple form is dead; what replaces it is a two-stage model in which starlight supplies a small bias and water on an asteroid amplifies it.
Established The pristine returned samples are the strongest single result on this page, and they point the other way. Analysis of material returned from asteroid (101955) Bennu identified 33 amino acids including 14 of the 20 used in terrestrial protein synthesis, with ammonia at 13.6 µmol g−1 in hot-water extract. Established Every chiral non-protein amino acid that could be enantiomerically resolved — isovaline, norvaline, beta-amino-n-butyric acid, beta-aminoisobutyric acid and 3-aminopentanoic acid — was racemic or near-racemic within analytical uncertainty. Established An L-valine excess of about 34% was measured after acid hydrolysis and was accompanied by elevated L-valine in the procedural blank, which is how a contamination signal looks when a laboratory is being careful about it. Established The authors state the consequence directly: the result challenges the hypothesis that left-handed protein-based life on Earth was influenced by an early Solar System bias toward L-amino acids.
Established Ryugu agrees. Hayabusa2 samples A0106 and C0107 gave total amino acid abundances of 8.3 ± 0.1 and 27.3 ± 1.9 nmol g−1 in unhydrolysed hot-water extracts, with alanine racemic within error in both — D/L of 1.09 ± 0.15 — and beta-aminobutyric acid racemic in every fraction analysed. Handwave The racemic reading of Ryugu is not uncontested — a 2024 Europlanet conference abstract is titled as a challenge to it, on the basis of an enantioselective re-analysis of the Orgueil meteorite — but that abstract could not be retrieved for this brief and is cited on its title alone, which is worth stating because a title is not a result.
Established On the sugar side there is no extraterrestrial chirality data at all. Bennu samples contain ribose at 0.097 ± 0.014 nmol g−1, glucose at 0.35 ± 0.05, plus arabinose, xylose, lyxose and galactose. Established The authors state that enantiomeric and isotopic measurement of those sugars was not possible, because of low abundance, interfering species and the sample mass available. Frontier Half of biology’s homochirality problem — the D-sugars — has never been measured in an extraterrestrial sample, and the limiting quantity is milligrams of returned rock.
Established The Soai reaction is the field’s existence proof and it is a narrow one. Diisopropylzinc adding to pyrimidine-5-carbaldehyde produces a pyrimidyl alkanol that catalyses its own formation with the same handedness, and the amplification is extreme: 0.00005% to above 99.5% in three rounds. Established Almost any chiral influence initiates it: d-quartz gives 97% enantiomeric excess of the S product and l-quartz 97% of the R, left-circularly-polarised light at 313 nm gives S product above 99.5%, and the reaction responds to isotopic chirality alone, with 12C/13C, 14N/15N, 16O/18O and H/D isotopomers all serving as initiators. A reaction that can read a carbon isotope substitution as a chiral instruction is a remarkable piece of physical organic chemistry and a poor model for prebiotic Earth, because nothing about it is prebiotic.
Established The statistical experiment matters more than the amplification one. Running the reaction 129 times under solid-vapour conditions with no chiral factor present produced the R alkanol 61 times and the S alkanol 58 times, with 10 trials below the 0.5% detection threshold. Established That is spontaneous absolute asymmetric synthesis with a fair coin, and it is the cleanest laboratory demonstration that a system can break mirror symmetry without any chiral input. Speculative It also demonstrates the contingent answer rather than the deterministic one, which is the opposite of what the parity-violation programme is looking for.
Frontier For twenty-five years the Soai reaction had no relatives outside its discoverers’ laboratory, and in 2020 it acquired one. Athavale, Simon, Houk and Denmark reported asymmetry-amplifying autocatalysis in the alkylation of 5-(trimethylsilylethynyl)pyridine-3-carbaldehyde with diisopropylzinc, and resolved the mechanism as a tetrameric zinc alkoxide catalyst operating through what they call a pyridine-assisted cube escape, with two-point substrate binding and intra-complex alkyl transfer. Frontier A second system found by a group that did not discover the first is the single most useful development in this subject since 2003, because it moves the phenomenon from anecdote toward class. Speculative It is still organozinc chemistry in an aprotic solvent, and it still has no water in it.
Established Circularly polarised photolysis has a measured ceiling, and it is low. Synchrotron circular dichroism of thin isovaline films across 130 to 330 nm resolves four anisotropy bands, with g173 = −0.0035 and g192 at least 0.0012. Established The maximum enantiomeric excess those anisotropies permit under complete photolysis is about 1.6%, and direct irradiation at 192 nm produced helicity-dependent excesses of up to about 2%, in two replicates per handedness. Frontier Asymmetric photolysis is real, reproducible, and about one and a half orders of magnitude short of the meteoritic excesses it is invoked to explain, which is why the current model puts aqueous alteration downstream of it rather than beside it. Speculative The astronomical input is circumstantial: near-infrared circular polarimetry finds large areas of circularly polarised light in the Orion and NGC 6334V star-forming regions, produced by scattering off aligned grains and by dichroic extinction, but the measurements are in the near infrared and the photochemistry is in the vacuum ultraviolet.
Frontier The magnetic-surface route is the most quantitatively impressive recent result and the most recently complicated. Crystallising racemic ribose-aminooxazoline, an RNA precursor in the cyanosulfidic route, on a magnetised magnetite surface gave about 60% enantiomeric excess in one step through the chiral-induced spin selectivity effect, and a subsequent crystallisation gave homochiral crystals at 100%. Frontier The reverse effect was demonstrated in the same year: crystallising enantiopure ribose-aminooxazoline onto an unmagnetised magnetic surface magnetises it, with an estimated effective field of about 2.5 mT, roughly 50 times the modern geomagnetic field, and a coercivity increase of about 20 times, from 75 mM solutions crystallised overnight at room temperature. Speculative Together those two results describe a feedback loop, and a feedback loop is what an origin-of-homochirality mechanism has to be.
Frontier The 2026 characterisation of prebiotically plausible magnetite is where that story gets harder. Magnetite synthesised by ultraviolet photo-oxidation and by nitrite-mediated oxidation of Fe(II) is dominated by single-vortex and multi-vortex magnetic domain states, and differs markedly from the nano-fabricated thin films every previous chiral-induced spin selectivity experiment used. Speculative Three-dimensional micromagnetic simulation says such grains undergo irreversible exchange-driven re-magnetisation on contact with spin-polarised homochiral molecules for exchange energies of 0.1 to 30 meV at particle sizes of 20 to 130 nm, and that vortex-state particles above about 100 nm have magnetic relaxation times exceeding the age of the Solar System. Handwave None of that is a measurement of the exchange energy between a real chiral molecule and a real magnetite grain, which the authors say has not been made; the simulation uses estimates spanning 0.01 to 100 meV and calls them conservative.
Established Parity violation is the only deterministic candidate and it has never been detected in a molecule. The parity-violating energy difference between enantiomers is calculated at about 100 aeV to 1 feV, equivalently 10−11 to 10−10 J mol−1, depending on the molecule. Established In frequency terms the target is brutal: the predicted parity-violating shift of the carbon–fluorine stretch in CHFClBr is about 2.4 mHz, a relative shift of about 8 × 10−17. Established The best experimental bound is ΔνPV < 8 Hz on CHFClBr, from a 1999 measurement at an experimental sensitivity of 2 × 10−13. Frontier That is a gap of about three and a half orders of magnitude between what has been excluded and what is predicted, and it has not moved in a quarter of a century.
Frontier Two developments have narrowed it from the theory side rather than the instrument side, and this is where the commission’s decisive experiment needs correcting. The commission predicts the parity-violating energy difference “at 10−17 to 10−15 of kT”. Established Taking the reviewed range of 10−11 to 10−10 J mol−1 against kT at 298 K, which is 2.48 kJ mol−1, gives 4 × 10−15 to 4 × 10−14 of kT — one to three orders of magnitude larger than the commission states. Frontier The figure the commission quotes is the right order for a light biological molecule such as alanine, where the roughly fifth-power dependence on nuclear charge suppresses the effect; it is the wrong order for the heavy-atom molecule the same sentence asks the experiment to use. Molecular candidates have moved further still: a 2024 quantum-dynamics study of substituted chiral methanols predicts parity-violating shifts of 186.4 mHz for a torsional transition in CFClBrOH and up to 3.1 Hz in CHBrIOH, against 1.867 mHz for the conventional carbon–fluorine stretch in CHFClBr. Frontier A predicted 3.1 Hz shift sits within a factor of three of the twenty-five-year-old experimental upper limit and two orders of magnitude above the sensitivity a planned two-photon Ramsey interferometer is designed to reach.
Frontier A second route is being opened in solution, and it is much cheaper. Chiral co-sensing measures diastereomeric splittings for a light and a heavy nucleus simultaneously in nuclear magnetic resonance, so that sample-preparation and drift errors largely cancel and effects below the linewidth become accessible; the reported residual 31P splitting at the racemic point was −56 ± 61 mHz, against a computed parity-violating splitting of −0.7 µHz at 20 tesla. Two to three orders of magnitude short, in a technique whose systematics are understood.
4 · Frontier questions
Frontier Six questions carry this subject, and they are ordered here by what a result would settle rather than by how much attention they get.
Speculative One. Is the sign determined or drawn? This is the only question on the list with a clean yes/no answer and a laboratory route to it. Frontier If the parity-violating energy difference is measured and its sign favours L-amino acids and D-sugars, the deterministic account becomes testable in a way it has never been; if it is measured and favours the wrong hand, the deterministic account is finished. Handwave And if it is measured at the predicted magnitude and favours the right hand, that still does not show it caused anything, because 10−14 of kT does not by itself select a molecule out of a racemate.
Frontier Two. Did the bias arrive or arise? The returned-sample data have changed the answer within five years. Established Every resolvable chiral non-protein amino acid in Bennu is racemic, and alanine in Ryugu is racemic at D/L 1.09 ± 0.15, while altered meteorites carry 10 to 20% L-isovaline. Frontier The most pristine material has the least excess, which means the excess is made where the water is, and the delivery hypothesis has become a hypothesis about parent-body chemistry rather than about impacts. Speculative Whether the same alteration chemistry ran on early Earth, where the water was, is untested.
Speculative Three. Is amplification one step or a ladder, and does anything climb it unattended? Every demonstrated amplifier has a discrete-step character: resubmit the Soai batch three times, recrystallise the aminooxazoline twice, grind the conglomerate for days. Handwave A geological setting that performs those operations is easy to narrate and has never been instrumented, and narration is doing more work in this literature than anyone admits.
Frontier Four. What is the abiotic ceiling on enantiomeric excess? This is the question the biosignature community needs answered and the origins community has no incentive to answer; the measured values give a working ceiling near 20% for natural samples and near 100% for engineered ones. Speculative Nobody has run the survey that would turn that into a distribution with a tail, which is the only form in which the number is usable as a detection threshold.
Speculative Five. Does handedness transfer between molecular classes? Life needs L-amino acids and D-sugars together, and a mechanism that enriches one says nothing about the other unless coupling is demonstrated. Frontier The magnetite work enriches an RNA precursor; the photolysis work enriches an amino acid; no experiment has shown an enriched pool of one class producing an enriched pool of the other. Handwave The 2026 magnetite paper lists exactly this as unproven in its own discussion, which is the correct place for it to appear.
Speculative Six. Is homochirality a precondition for heredity or a consequence of it? The standard argument is enantiomeric cross-inhibition, demonstrated in poly(C)-directed oligo(G) synthesis in 1984: the wrong-handed monomer binds, terminates the chain and poisons the reaction, so a racemic pool cannot make long polymers. Handwave Whether that is a barrier or a selection pressure is unresolved — a system that can only polymerise from an enriched pool also rewards any enrichment — and nobody has built the experiment that distinguishes the two readings.
5 · Technological bottlenecks
Established The first bottleneck is three and a half orders of magnitude wide and is the whole parity-violation programme. The best bound is 8 Hz; the conventional prediction is 2.4 mHz. Frontier Closing it needs spectroscopic resolution at the 0.01 Hz level on a molecule that is chiral, volatile, thermally stable, resolvable into enantiomers in usable quantity, and contains a heavy nucleus — and those requirements fight each other, which is why the field has spent two decades on a small number of candidate molecules rather than a large one. Speculative The 2024 large-amplitude-motion result changes the trade: it finds hertz-scale shifts in molecules whose heaviest atoms are bromine and iodine, which relaxes the heavy-nucleus requirement and replaces it with a synthesis problem.
Frontier The second is the contamination floor, and it is the reason this subject’s numbers are so hard to compare. A 34% L-valine excess measured in Bennu material turned out to be tracked by the procedural blank. Established That is a well-run laboratory catching its own artefact, and it is also a statement about the size of the effect the field is chasing: the interesting excesses and the contamination excesses are the same size. Speculative Without a blank of comparable rigour, any reported enantiomeric excess in the 10 to 40% range in a returned or terrestrial sample is uninterpretable, and most of the older literature does not report one.
Frontier The third is sample mass. Bennu’s sugars are present at tens to hundreds of picomoles per gram, and enantiomeric analysis was not possible at that abundance with the material available. Speculative Chiral separation costs sample in a way achiral identification does not, because the analyte is split between two peaks and the derivatisation adds steps. Handwave The quantity that would fix this is grams of pristine carbonaceous asteroid, and the missions that return it are decadal.
Frontier The fourth is that the magnetic route’s demonstrations used the wrong magnetite. Every chiral-induced spin selectivity crystallisation result to date used nano-fabricated thin films; magnetite formed by plausible prebiotic routes is dominated by single-vortex and multi-vortex domain states with different magnetic behaviour. Speculative The 2026 simulation argues that this is favourable rather than fatal, because vortex-state grains re-magnetise irreversibly and hold that state geologically. That argument is a micromagnetic calculation with an unmeasured exchange energy in it.
Handwave The fifth is not technical and is the most damaging: there is no negative control. The field has no published statement of what an abiotic system cannot do with enantiomeric excess, which means no result can fail. Speculative A mechanism that produces 2% is reported as symmetry breaking, one that produces 60% is reported as amplification, and one that produces 100% is reported as homochirality; none is reported as insufficient, because insufficiency has never been defined. Frontier The comparison with the biosignature community is instructive, because that community has at least written its confidence scale down.
6 · Research dependencies
Frontier This subject depends on five other fields and contributes to none of them, which is a fair description of its funding position.
Established Relativistic quantum chemistry sets every target the parity-violation experiments aim at. The predicted shifts — 2.4 mHz for a carbon–fluorine stretch, hundreds of millihertz for methyltrioxorhenium complexes, 3.1 Hz for a torsional mode in CHBrIOH — come from four-component and exact two-component electronic-structure calculations coupled to nuclear dynamics. Frontier An experiment that finds nothing where the theory says to look is ambiguous between instrument and calculation, and this is the field where that ambiguity bites hardest.
Frontier Cold and ultracold molecular physics supplies the coherence times. The Ramsey-interferometry approach needs a continuous supersonic jet of a chiral species with a usable mid-infrared transition, and the emerging ion-trap approaches need chiral molecular ions that can be state-prepared. Speculative Neither community has chirality as its own objective; both are pursuing electron-dipole-moment and time-reversal-violation searches, and chiral molecules arrive as a byproduct of that programme.
Established Analytical separation science sets the floor on every meteoritic number. The current results rest on ultra-high-performance liquid chromatography with fluorescence detection and quadrupole time-of-flight mass spectrometry, and on gas chromatography coupled to isotope-ratio mass spectrometry for the carbon-isotope confirmation that a compound is extraterrestrial. Frontier The isotope measurement is the load-bearing one for chirality claims, because it is the only thing that distinguishes an indigenous excess from a contaminating one.
Frontier Rock and environmental magnetism supplies the prebiotic-magnetite question. Domain-state determination, coercivity spectra and three-dimensional micromagnetic modelling are palaeomagnetism techniques, and the 2026 paper is a palaeomagnetism paper wearing an origins-of-life title. Speculative That is a genuinely new dependency, created in the last three years by the chiral-induced spin selectivity results.
Frontier And sample return supplies the only uncontaminated material. The Bennu and Ryugu results exist because two agencies built curation chains that keep terrestrial organics out, and the value of those chains to this question was not the reason they were built. Speculative The next increment in this subject’s evidence base is a mission, not an experiment, and no mission is currently scoped to deliver enantiomeric analysis of extraterrestrial sugars.
7 · Required experiments
Frontier Every position in section 4 has a test, and they rank by what a result would settle rather than by how buildable they are.
Speculative Tier one, and the decisive experiment for this brief: measure the parity-violating frequency difference between two enantiomers of a heavy-atom chiral molecule at the 10 mHz level. This is the only experiment on the list that separates a determined universe from a drawn one, and it is a measurement rather than a demonstration. Established The target has a number: about 2.4 mHz for the carbon–fluorine stretch of CHFClBr, a relative shift near 8 × 10−17, against the standing experimental bound of 8 Hz set in 1999 at a sensitivity of 2 × 10−13. Frontier The two-photon Ramsey-interferometry approach on a continuous supersonic jet is designed for a few times 0.01 Hz, which is sufficient for the enhanced candidates and insufficient for CHFClBr by two orders of magnitude. Speculative The best current candidate is a torsional transition in CHBrIOH at a predicted 3.1 Hz, because a predicted shift larger than the projected sensitivity by two orders of magnitude turns a heroic measurement into an ordinary one. Handwave Nobody has synthesised and enantiomerically resolved that molecule in jet-compatible quantity, and the paper proposing it is a calculation.
Frontier Tier one-and-a-half, because it is running now and nobody expects it to succeed first: the solution-state route. Chiral co-sensing in nuclear magnetic resonance reached a residual 31P splitting of −56 ± 61 mHz against a predicted parity-violating splitting of −0.7 µHz at 20 tesla. Speculative Two to three orders short, with the dominant systematics already identified and cancelled by design, in an apparatus that exists in hundreds of chemistry departments. Frontier It is the cheapest serious attempt on this list by a wide margin.
Speculative Tier two is the experiment the field most needs and least wants: the abiotic ceiling survey. Take every mechanism known to generate enantiomeric excess without biology — asymmetric photolysis, mineral-surface adsorption, magnetised-surface crystallisation, eutectic enrichment, attrition-driven deracemisation, spontaneous conglomerate formation — and measure the distribution of excesses each produces across a wide range of substrates and conditions, reporting the tail rather than the best case. Frontier The output is a threshold: the enantiomeric excess above which no abiotic mechanism has been observed to reach, with a stated false-positive rate. Handwave Nobody funds a survey whose product is a number that makes other people’s claims harder to publish, and that is the honest reason this has not been done.
Speculative Tier three is the end-to-end magnetite run under conditions the 2026 paper specifies. Synthesise magnetite by ultraviolet photo-oxidation or nitrite-mediated oxidation of Fe(II), magnetise it in a field of geomagnetic order, crystallise racemic ribose-aminooxazoline on it, measure the enantiomeric excess, measure the resulting grain magnetisation, and repeat the cycle without re-magnetising by hand. Frontier The feedback loop is the claim; no experiment has yet closed it, and the two halves were demonstrated separately on thin films. Speculative The measurable that decides it is whether the excess grows over cycles or saturates, and the paper’s own unproven list names the exchange energy between a real chiral molecule and a real magnetite grain as the quantity nobody has measured.
Speculative Tier four is a chirality-transfer experiment, and it is the one nobody is running. Take a pool of ribose-aminooxazoline at 60% enantiomeric excess, run it forward through a published prebiotic nucleotide synthesis, and measure the enantiomeric excess of the product and of any co-produced amino acid. Frontier A demonstration that handedness survives a synthetic step at all would be the first evidence that a local enrichment can become a systemic one. Handwave The reason it has not been run is that the prebiotic syntheses it would run through are themselves multi-stage sequences with a chemist between the stages, which is the problem Prebiotic Chemical Networks owns.
Frontier Tier five is a sample-return measurement with a stated mass requirement. Enantiomeric analysis of extraterrestrial sugars has never been performed, and the stated obstacle is abundance, interference and available mass rather than technique. Speculative The experiment is a sugar enantiomer measurement on a pristine carbonaceous sample, and the specification a mission would have to meet is the milligram-to-gram figure nobody has published as a requirement. Established The amino-acid half of that measurement has already been made twice, on Bennu and Ryugu, and both came back racemic.
Frontier Tier six is replication of the 2020 second autocatalyst, and it is cheap. A second asymmetry-amplifying autocatalytic system found outside the discovering laboratory converts a singular curiosity into a class with at least two members. Speculative A systematic search for more, guided by the tetrameric-cluster mechanism rather than by resemblance to the original substrate, is a tractable programme with a clear negative result available. Handwave Whether any member of that class can be made to work in water is the question that decides whether the whole line is relevant to prebiotic chemistry, and no one has reported a water-compatible example.
Established And the experiments that are not tests of this and get reported as though they were. Industrial asymmetric catalysis, enantioselective organocatalysis and chiral chromatography are measurements of how well chemists can control chirality with chiral reagents in hand. Speculative They are irrelevant to the origin question by construction, because every one of them starts with a chiral input, and the origin question is about where the first chiral input came from. Frontier The Soai reaction is the exception that proves the rule, and only in its 129-trial no-chiral-input form.
8 · Engineering requirements
Frontier Four instruments carry this subject, and only one of them has been built for it.
Speculative The parity-violation spectrometer is the demanding one. A two-photon Ramsey interferometer on a continuous supersonic jet, with an ultra-stable mid-infrared source referenced to a frequency comb, targeting a few times 0.01 Hz on a molecular transition near 10 µm, with the two enantiomers measured under conditions identical to the part in 1013. Frontier The systematic that dominates is not statistical sensitivity but the difference between the two samples: any impurity, pressure or field asymmetry between the L and D runs appears as a parity-violating signal. Speculative The solution-state co-sensing approach is engineering around exactly that, by measuring two nuclei in the same tube so the asymmetry cancels.
Frontier The flight instrument is the one with a customer. Enantiomeric analysis of extraterrestrial organics requires chiral separation combined with an ionisation path that does not destroy the stereocentre, and no such instrument has ever operated off Earth. Speculative The requirements a mission would have to close are sample mass, derivatisation chemistry that survives launch and storage, and an organic blank good enough that a 10% excess is interpretable. Frontier Biosignature Standards carries the argument about what such an instrument’s output would be worth; this page supplies the number it would have to beat.
Established The magnetic route needs materials engineering more than physics. Producing magnetite with a controlled distribution of single-domain, single-vortex and multi-vortex grains, characterising it by electron microscopy and first-order reversal curves, and then running enantioselective crystallisation on the powder rather than on a film. Speculative The measurement that has never been made is a direct determination of the exchange energy between an adsorbed chiral molecule and a magnetite surface, which the 2026 simulation treats as a free parameter spanning four orders of magnitude.
Frontier And the cheapest requirement is not an instrument at all. A shared reference set of racemic and enriched standards, with agreed derivatisation and blank protocols, would make the numbers in this subject comparable between laboratories. Handwave It does not exist, it would cost very little, and its absence is why a 37% alanine excess from one laboratory and a 10% isovaline excess from another cannot be placed on the same axis.
9 · Adjacent technologies
Established The nearest page on this map is Prebiotic Chemical Networks, and the division of labour is clean. That brief owns the heredity problem — yields, replicator fidelity, whether a network reaches a self-sustaining state — and treats the racemic composition of Bennu as one entry in its misconceptions list. Frontier This page owns the chirality axis: which hand, how it was chosen, and what the measured ceiling on an abiotic excess is. The two meet at one point, which is that the best prebiotic syntheses are multi-stage sequences with a chemist between the stages, and a chirality-transfer experiment inherits that problem whole.
Frontier The commission named Xenobiology as the nearest existing brief, and it is adjacent on a different axis. That page is about orthogonality — alternative backbones, expanded alphabets, recoded genomes, and the mirror-life moratorium argument — and it identifies chiral analytical chemistry at environmental sensitivity as a dependency it needs and nobody is building. Speculative This page supplies the baseline that dependency requires: what enantiomeric excesses occur naturally, and therefore what an anomaly would have to exceed. Frontier The two pages are complementary rather than overlapping, because a mirror organism is an engineering object and this brief is about a historical event.
Frontier Biosignature Standards is the page that consumes this one’s output. Enantiomeric excess is the agnostic biosignature with the best ratio of diagnostic power to ambiguity, and its diagnostic power is set by the abiotic ceiling that section 5 says nobody has measured properly. Speculative A confidence scale for life detection cannot grade a chirality claim without that number, and the number is a chemistry result rather than a mission result.
Speculative Three further neighbours inherit something specific. Assembly Theory is the sibling case of an agnostic, substrate-independent life-detection proposal whose threshold is contested, and the two briefs share the methodological problem of a missing abiotic baseline. Precision Quantum Sensing builds the frequency references and cold-molecule sources the parity-violation measurement needs, and gains nothing back. Planetary Protection inherits the contamination problem in its most acute form, because a chirality measurement on another body is exactly the measurement terrestrial organics ruin.
Frontier One adjacency is economic rather than scientific. The parity-violation measurement rides on apparatus built for electron-electric-dipole-moment and time-reversal-violation searches, and the meteoritic measurements ride on curation chains built for cosmochemistry. This subject owns almost none of the instruments it depends on, which is the same business model Laboratory Astrophysics runs.
10 · Institutional requirements
Established The institutional facts that matter here are about curation, blanks and who is allowed to say no.
Established Sample-return curation is the one institution this subject depends on absolutely and does not control. The Bennu and Ryugu chirality results exist because two agencies maintained contamination-controlled chains from collection through allocation, and because the analysing laboratories ran procedural blanks good enough to catch a 34% L-valine artefact. Frontier That discipline is expensive, invisible in the resulting papers, and the reason the racemic result is believable.
Speculative Allocation is a real constraint and is decided elsewhere. Enantiomeric analysis of Bennu’s sugars failed on sample mass, not on technique, and returned mass is allocated by committees balancing many disciplines against a fixed quantity of rock; a chirality measurement competes with mineralogy and isotope work for the same milligrams, and it is the destructive option.
Speculative The parity-violation programme has an institutional problem of a different kind: it is a twenty-five-year measurement with no interim deliverable. The standing bound dates from 1999 and the field’s publications since are calculations, proposals and apparatus papers. Frontier That is a funding profile almost no agency runs, and the work survives because it is parasitic on the fundamental-symmetry-violation programme, which has its own justification.
Handwave The cheapest institutional intervention available is a convention nobody has written. An agreed statement of what counts as an abiotic enantiomeric excess, with a reference standard set and a blank protocol, would cost a workshop and a small consortium. Speculative Its absence is why the numbers in section 3 come with method footnotes rather than error bars that mean the same thing, and why a 2024 conference abstract can contest a returned-sample result without anyone being able to adjudicate it quickly.
Frontier And the field has no home. Homochirality sits between cosmochemistry, physical organic chemistry, molecular physics and astrobiology, and is nobody’s programme line. Speculative That is why the strongest results on this page were produced as byproducts — of a dark-matter-adjacent symmetry programme, of an asteroid mission, of a synthetic chemistry group’s curiosity — and why the survey section 7 asks for has no natural sponsor.
11 · Ethical & societal considerations
Frontier The ethical weight here is mostly epistemic, and the exception is large.
Established The exception is mirror life, and this page defers it. A biosphere built on D-amino acids and L-sugars is the direct engineering consequence of understanding chirality well enough to invert it, and the December 2024 statement asking that mirror bacteria never be built is handled where it belongs, in Xenobiology. Speculative What this page adds is that the origin question and the mirror-life question are not the same question, and conflating them has a cost: a moratorium argued from the risk of mirror organisms should not chill measurement of a natural enantiomeric excess in a rock.
Frontier The routine ethical problem is overclaiming, and chirality is unusually prone to it. An enantiomeric excess is a single number with an obvious narrative attached, and the narrative — life leaves fingerprints — is more compelling than the statistics usually support. Established The 34% L-valine excess in Bennu material would have been a headline if the blank had not been run. Speculative The discipline that prevents the next version of that is a published abiotic ceiling, which is a scientific product with an ethical function.
Frontier There is a real and under-discussed issue about destructive analysis of irreplaceable material. Returned asteroid samples are finite, chirality measurement consumes them, and the measurement cannot currently be made non-destructively. Speculative Choosing to spend Bennu on a chirality answer is choosing not to spend it on something else, and that trade is made by allocation committees rather than in public.
Speculative The pharmaceutical framing that attaches itself to this subject is mostly a distraction and occasionally a misrepresentation. Single-enantiomer drug manufacture is a solved industrial problem using chiral catalysts and chiral separations, and it does not wait on an origin-of-homochirality result. Handwave A grant application that motivates parity-violation spectroscopy by thalidomide is describing a connection that does not carry weight in either direction.
Frontier And there is the question of what a null does to a field. If the parity-violating energy difference is measured and turns out to favour the wrong hand, the deterministic account ends and a large literature becomes history. Speculative A field that has spent decades on a measurement has an interest in the measurement being ambiguous, and the standard protection against that is preregistration of what each outcome means — which nobody in this subject has done.
12 · Civilizational implications
Speculative The civilizational content of this question is one bit, and which bit it is matters more than most single bits in science.
Speculative If handedness is determined, life is more predictable than the contingency argument allows, and the sign of an extraterrestrial excess carries no information about independent origin. A biosphere found on an ocean moon with L-amino acids would be consistent with a second genesis and with panspermia, and the measurement that would have distinguished them would have been thrown away before the mission launched. Frontier That is a concrete, expensive consequence of not having made a laboratory measurement.
Speculative If handedness is drawn, the sign is a fingerprint. A mirrored biosphere is then proof of an independent origin at close to the strength of a direct observation, which is a rare thing in astrobiology and would be the single most consequential result the discipline could produce. Handwave A same-handed biosphere would be ambiguous in the other direction and would push the panspermia question back into the foreground, where it has no experimental traction.
Speculative There is a third outcome that gets less attention and may be the likeliest: mixed. A deterministic bias too small to matter, plus a contingent amplifier that happened to run in one direction, gives a universe in which handedness is weakly predictable and locally accidental. Handwave That answer is hard to publish, hard to test, and is what the current numbers most naturally suggest — a 10−14-of-kT energy difference is not going to select a molecule out of a racemate on its own.
Frontier The near-term civilizational consequence is smaller and more definite: it is about instrument choice. Chirality-capable separation is a specific payload decision that competes with mass spectrometry, imaging and drilling for a fixed budget. Speculative The case for flying it is exactly as strong as the abiotic ceiling is well characterised, which means a laboratory survey on Earth is on the critical path of a flagship mission’s payload manifest.
Handwave Claims that solving homochirality would enable anything — new materials, new pharmaceuticals, new computation — run in the wrong direction and are not made here. The chemistry that would follow from the answer is chemistry we already have; what would follow is a settled fact about history.
13 · Timelines
These horizons track measurement sensitivity and sample availability, because there is no engineering programme in this subject to track:
- 10 yr: Frontier Expect the parity-violation bound to move for the first time since 1999, from Ramsey interferometry or from solution-state co-sensing, and expect the first move to be a factor of ten to a hundred rather than the three and a half orders of magnitude needed. Speculative Expect the magnetite route to be tested on prebiotically plausible powders rather than thin films, with a real chance that the effect is smaller than the thin-film results imply. Frontier Expect at least one more asteroid or comet sample with amino-acid chirality data, and expect it to be racemic.
- 25 yr: Speculative This is the window in which the decisive measurement plausibly lands: a parity-violating frequency difference resolved at the 10 mHz level in a molecule chosen for a large-amplitude-motion enhancement rather than for a heavy nucleus. Speculative A positive result with the sign favouring L-amino acids reopens every question in section 4 and makes the abiotic-ceiling survey urgent; a positive result with the wrong sign closes the deterministic account permanently. Handwave A null at the predicted magnitude would be the least informative outcome and the most likely, because it is consistent with the calculation being wrong.
- 50 yr: Speculative On the chemistry side the plausible development is an end-to-end demonstration in one vessel — racemic feedstock to enriched polymer without intervention — or the recognition that the hand-off between stages is the phenomenon rather than an inconvenience. Handwave On the observational side, an enantiomeric measurement on material from an ocean moon is the only thing that would settle the contingency question empirically, and it requires a mission class that does not currently exist.
- 100 / 250+ yr: Handwave Beyond useful forecasting. The defensible statement is structural: a question with one laboratory route and one observational route either gets answered by one of them or stops being asked. Handwave Any specific claim about what a confirmed deterministic bias would be used for is an assertion, and this page does not make one.
14 · Technology tree & dependencies
- Depends on One typed dependency on this map and it is soft: Prebiotic Chemical Networks supplies the syntheses a chirality-transfer experiment would have to run through, and its finding that no prebiotic route has a published end-to-end carbon yield is the reason the transfer experiment in section 7 cannot currently be specified. Precision Quantum Sensing supplies the frequency references and cold-molecule technique the decisive measurement needs and is not blocking, because that hardware is being built for metrology and symmetry tests whether or not anyone measures a chiral molecule with it. Nothing else on this map gates this subject.
- Requires (not on this map) Six conditions, in the order of the tokens above. The measurement. A parity-violating frequency difference resolved at the 10 mHz level between two enantiomers of a heavy-atom or large-amplitude-motion chiral molecule; the standing bound is 8 Hz and the conventional prediction is 2.4 mHz, so the gap is three and a half orders of magnitude and has not moved since 1999. The ceiling. A measured distribution, not a best case, of the enantiomeric excess each known abiotic mechanism produces, with a stated tail — the number that turns chirality into a usable detection threshold and the number nobody is paid to produce. The flight instrument. Chiral separation with a non-destructive ionisation path, qualified for planetary flight; mature in terrestrial laboratories, never operated on another body. The sample. Enough pristine carbonaceous material to resolve sugar enantiomers, which failed on Bennu at ribose abundances near 0.1 nmol g−1. The route. One vessel, racemic in, enantioenriched polymer out, no chemist between stages — the requirement every demonstrated amplifier currently fails. The protocol. An agreed blank and reference-standard convention so that excesses reported by different laboratories can be placed on one axis; the cheapest item on this list and the one whose absence does most damage.
- Enables A measured parity-violating energy difference would make the sign of an extraterrestrial enantiomeric excess interpretable, which is the input Biosignature Standards needs to grade a chirality claim above its lowest confidence levels, and would give Xenobiology a quantitative statement about whether a mirror biochemistry is disfavoured by physics or merely unbuilt. No typed enabling edge is claimed beyond those, because a question that has never been answered enables nothing on a schedule.
- Adjacent Cosmochemistry and sample-return curation, physical organic chemistry, molecular and cold-atom physics, and rock magnetism supply every number on this page. Within this map: Prebiotic Chemical Networks for the heredity problem this one sits beside, Assembly Theory for the sibling problem of an agnostic life-detection threshold with a missing abiotic baseline, Xenobiology for mirror chemistry as an engineering object, Biosignature Standards for what a chirality claim is worth, Planetary Protection for the contamination regime, and Laboratory Astrophysics for the parasitic-instrument economics.
15 · Common misconceptions & speculative claims
Established “Meteorites delivered the left-handed bias, and that is why life is left-handed.” The most pristine extraterrestrial material ever returned is racemic. Every chiral non-protein amino acid resolvable in Bennu samples came back racemic or near-racemic within uncertainty, and alanine in Ryugu came back at D/L 1.09 ± 0.15. Frontier The hypothesis is not dead but it has moved: the excesses are made by aqueous alteration on the parent body, not delivered from the interstellar medium, which is the explicit conclusion of the 2009 alteration study and is consistent with pristine meteorites being racemic and altered ones enriched. Prebiotic Chemical Networks reaches the same conclusion from the feedstock side; this page adds that the alteration model makes the excess a local product of water chemistry, which is a claim about early Earth as much as about asteroids.
Established “The Soai reaction shows how homochirality arose.” It shows that asymmetric autocatalysis with extreme amplification is chemically possible, which was not obvious before 1990 and is a real result. Frontier It does not show that it arose this way, because the reaction is diisopropylzinc adding to a pyrimidine aldehyde in an aprotic solvent, water destroys it, and no prebiotically plausible analogue has been reported in three decades. Established The amplification itself is also routinely misdescribed: 0.00005% to above 99.5% took three consecutive rounds, and the discovering group describes it that way. Frontier The strongest honest reading is the 2019 statistics — 129 trials with no chiral input giving 61 R and 58 S — which demonstrates spontaneous symmetry breaking and, in doing so, argues for contingency rather than for a determined sign.
Established “Parity violation explains homochirality.” Parity violation is the only mechanism that could explain it universally, and it has never been observed in a molecule. Established The predicted energy difference is about 10−11 to 10−10 J mol−1, which is roughly 10−14 of kT at room temperature for a favourable heavy-atom molecule and smaller still for an amino acid. Speculative An energy bias that small does not select a molecule out of a racemate; it can only bias a system that is already amplifying, and it would need the amplifier to be sensitive at that level, which no amplifier has been shown to be. Handwave The step where the argument works by assertion is always the same one: that a sufficiently sensitive autocatalytic network converts a 10−14 energy bias into a macroscopic excess, given enough time. Nobody has built that network.
Established “Circularly polarised starlight explains it.” Asymmetric photolysis works and its ceiling has been measured. Established The anisotropy spectrum of isovaline permits about 1.6% enantiomeric excess at complete photolysis, and irradiation at 192 nm delivered about 2%. Frontier Two per cent is an order of magnitude below the meteoritic excesses and two orders below biology, so circularly polarised light is a plausible initiator and a demonstrated non-solution to the amplification problem. Speculative The astronomical side is also thinner than the citation pattern suggests: the polarimetry showing large circularly polarised regions in Orion and NGC 6334V is near-infrared, and the photochemistry that matters is in the vacuum ultraviolet.
Frontier “The magnetite result solved it.” Crystallising an RNA precursor on a magnetised surface reached about 60% enantiomeric excess and then 100%, and a companion result showed the molecules magnetising the surface in return, which is a genuine feedback loop and the most quantitatively impressive thing in this subject. Speculative It used nano-fabricated thin films. The 2026 characterisation of magnetite made by plausible prebiotic routes finds single-vortex and multi-vortex grains that differ markedly from those films, and the argument that this is favourable is a micromagnetic simulation with an exchange energy estimated across four orders of magnitude. Handwave No experiment has yet closed the loop on prebiotic material, and the same paper lists chirality transfer between molecular classes as unproven.
Frontier “Homochirality is necessary for life.” It is necessary for the kind of polymer chemistry life uses, and the mechanism is enantiomeric cross-inhibition: in template-directed polymerisation the wrong-handed monomer incorporates and terminates the chain, which was demonstrated in poly(C)-directed oligo(G) synthesis in 1984. Speculative It does not follow that homochirality preceded life. A system that polymerises only from an enriched pool also rewards any enrichment, which makes cross-inhibition a candidate amplifier rather than a precondition, and no experiment distinguishes the two readings.
Frontier “An enantiomeric excess is a biosignature.” Not at the magnitudes abiotic chemistry reaches. Established Natural samples reach 10 to 20% without biology, and laboratory mechanisms reach 100%. Speculative The excess that would be diagnostic is one that exceeds every abiotic mechanism’s measured tail, and because that survey has not been run the threshold is set by convention rather than by data. Frontier Biosignature Standards makes the general version of this argument across all agnostic biosignatures; the chirality case is the one where the missing number is most specific and most cheaply obtainable.
Speculative “The answer will come from finding alien life.” It would, and it is the slower of the two routes by decades. Frontier The laboratory route — measuring the parity-violating energy difference — needs no mission, no launch window and no target, and has been technically conceivable since 1999. Handwave That it has not been done is a fact about funding and about molecular synthesis, not about physics.
Handwave “Homochirality is essentially solved; only details remain.” and “Homochirality is unsolvable because the evidence is gone.” Both are assertions and both are wrong in the same way, which is that they treat a three-part problem as one. Frontier Symmetry breaking is close to solved: several mechanisms make small excesses and one has been measured directly. Amplification is demonstrated in systems that are not prebiotic. Transfer between molecular classes has never been demonstrated at all, and that is the part where the evidence is thinnest and the experiments are cheapest.
