1 · Why this matters

Speculative If demixing itself is a control variable — if a cell changes what a set of molecules does by moving them from one phase into two, rather than by changing how tightly they bind — then there is a regulatory axis that occupancy pharmacology cannot see, and it is present in every compartment of every cell that has been looked at. The condition comes first because it has been demonstrated cleanly for very few proteins: section 7 says what a clean demonstration would look like, and section 5 says why almost nobody has produced one.

Established The magnitude, where it is measurable, is a concentration ratio and a material property, not a slogan. The Caenorhabditis elegans P-granule protein LAF-1 begins condensing at about 800 nM at 125 mM NaCl and forms droplets of viscosity 34 ± 5 Pa s, roughly honey; adding 5 µM RNA drops that to 12.8 ± 0.8 Pa s. Established A reconstituted RGG condensate measured by micropipette aspiration has a surface tension of 0.159 ± 0.010 mN/m, about 300 times lower than mineral oil, with a viscosity more than 200 times higher. Those are the units the prize is denominated in: a cell that can move a component across a threshold of a few hundred nanomolar can change the local chemistry of a micron-scale volume without building a membrane.

Frontier The financial version of the prize is one clinical asset and one closed company, and both numbers are known. Dewpoint Therapeutics had raised about $287 million across three rounds by February 2023, took a further Series D in September 2025, and dosed the first patient with DPTX3186, a small molecule that forces β-catenin into an inactive condensate, on 4 February 2026. Established Faze Medicines launched in December 2020 with $81 million from Third Rock, Novartis, Lilly and AbbVie, never entered the clinic, and shut in November 2022 because, in its lead investor’s words, the science did not progress enough to justify more money. The upside here accrues first to oncology, because that is where a condensate-modulating molecule can be paid for; the cell biology is universal and the capital is not.

Handwave The condition, stated plainly: every number above was measured in a tube or in a cell line expressing a tagged construct, and the step from “this protein demixes” to “this compartment works because it demixes” is the step the field has not generally taken. The one thing that would convert the prize from conditional to real is a mutation that removes the phase behaviour and nothing else, in a cell, with a function that fails only then — and section 5 explains why that mutation is hard to build rather than merely unfashionable.

2 · Concept overview

A biomolecular condensate is a compartment inside a cell that concentrates a specific set of molecules without a membrane around them. Nucleoli, Cajal bodies, stress granules, P granules, P-bodies and nuclear speckles are the canonical examples; all of them were described microscopically decades before anyone proposed a physics for them.

Established The physics proposed in 2009 was liquid–liquid phase separation, and the first evidence was mechanical rather than biochemical. P granules in the C. elegans germ line were shown to drip, fuse, wet surfaces and relax to spheres, with a measured viscosity of about 1 Pa s — roughly a thousand times water, comparable to glycerol — and a surface tension near 1 mN/m, about 10−5 of an air–water interface. Established Their asymmetric localisation in the embryo was explained not by transport but by dissolution at one end and condensation at the other, driven by a gradient in the saturation concentration.

Frontier The idea then generalised faster than the measurements did. Between 2009 and 2026 the same explanation was offered for transcription, heterochromatin, signalling, synaptic assembly, ribosome biogenesis, the innate immune response, and the aggregation that kills motor neurons. Speculative The generalisation may be correct. Handwave It is also exactly the shape a framework takes when it is being applied rather than tested, and that is why this page sits in Category XI rather than in the cell-biology categories.

Established The field has an inventory and it is not large. CD-CODE, the community condensate database published in Nature Methods in April 2023, held 244 distinct biomolecular condensates linked to 9,861 proteins across 49 organisms, plus 375 synthetic in vitro condensates, with 9,916 protein–condensate relationships each carrying a PubMed identifier as evidence. Frontier An inventory of a few hundred named compartments and ten thousand associated proteins is a real body of knowledge and a very long way from a mechanism established across the proteome.

Frontier This brief treats the in vitro record, the in-cell record, the causality problem, the material-state question, the therapeutic programme and the misconceptions. It does not treat tissue-scale self-organisation, which belongs to Embryo Models, and it does not treat the design of new binding proteins, which belongs to Generative Biomolecular Design. Established A note on sourcing: every measured number in this brief is attributed to a named paper in the reading list, and where the research for this brief could not retrieve a source it is cited by name with no link and the failure is stated.

3 · Current scientific position

Established The in vitro record is the strongest part of this subject and it is genuinely strong. Purified proteins demix from buffer at reproducible thresholds, the resulting droplets have measurable viscosity and surface tension, and the thresholds move with salt, temperature, RNA and post-translational modification in directions that polymer theory predicts. Established That is a real physical phenomenon, and no serious critic disputes it.

Established LAF-1 is the cleanest single case. At 125 mM NaCl it begins condensing at a critical protein concentration of roughly 800 nM; the droplets have a viscosity of 34 ± 5 Pa s, falling to 14 Pa s at 250 mM NaCl and 8 Pa s at 400 mM. Established Adding 5 µM RNA produces a threefold drop to 12.8 ± 0.8 Pa s, and the fluorescence-recovery timescale falls from about 233 s to about 94 s; LAF-1 binds RNA with a dissociation constant near 10 nM. Frontier The direction matters more than the magnitude: RNA makes this condensate more liquid, not less, which is the opposite of the intuition that more components means more gel.

Established FUS is the most-measured case and the most construct-dependent. A Nature Communications paper of November 2025 reports a saturation concentration of 0.5 to 1 µM for pre-cleaved FUS and 2 to 5 µM for tag-cleaved FUS — a factor of about five that comes from a purification decision rather than from the protein. Frontier The same work reports a distinct state below the saturation concentration: nanoclusters of 300 to 500 nm forming at about 200 nM, with arrested coarsening, equilibrium reached in roughly 30 minutes, and molecular exchange above 3,000 molecules per µm2 per second, comparable to a condensate. Speculative If the dilute phase is not dilute, the two-state picture that the whole field’s vocabulary rests on is an approximation, and the authors say the sub-saturated condition is the one commonly found in an unstressed cell.

Established The commission’s anchor named three proteins and it is two-thirds right. LAF-1 and FUS were both assigned saturation concentrations and material properties, as above. Established DDX4 was not. The 2015 study that made DDX4 a canonical condensate protein measured a transition temperature as a function of protein concentration and ionic strength, not a saturation concentration; its working condition was 100 µM of the N-terminal disordered region Ddx4N1, rising to 162.5 µM for the nucleic-acid partitioning experiments, and the construct called full-length in shorthand was Ddx4YFP, in which the helicase domain is replaced by a fluorescent protein. Established The material property reported was an internal diffusion coefficient of 4 ± 1 × 10−13 m2 s−1 in vitro and 3 ± 1 × 10−13 m2 s−1 in cells — not a viscosity and not a surface tension. Frontier The correction is not pedantry: 100 µM is about 125 times LAF-1’s threshold, so the three anchor proteins were not shown to demix in the same concentration regime, and a reader who takes “measured saturation concentrations” at face value will over-rate how comparable the three measurements are.

Established The one genuinely separating perturbation in that 2015 work is also its most cited result and it is not a separation-of-function mutant. Scrambling the charge pattern of Ddx4N1 while holding its amino-acid composition constant abolished organelle formation in vitro under near-physiological conditions and in cells. Frontier That establishes that the arrangement of residues, not merely their identity, sets the phase behaviour. Handwave It does not establish that phase behaviour rather than the altered interactions is what the compartment needed, because scrambling the charge blocks changes the interactions too — which is precisely the confound section 7 is about.

Established Condensate interiors have been shown to be chemically distinct, which is the mechanistic core of the whole idea. In the DDX4 system, single-stranded DNA was concentrated in the droplet interior while double-stranded DNA was largely excluded — a selectivity that is a property of the phase, not of a binding site. Frontier Selective partitioning is the best-supported functional consequence of condensation and it is still usually demonstrated in vitro.

Frontier Material properties are not one number, and the honest version of the record is a range across methods. Micropipette aspiration of an RGG condensate gives viscosity 1.62 ± 0.18 Pa s and surface tension 0.159 ± 0.010 mN/m over eleven droplets; P granules in an embryo gave about 1 Pa s and 1 mN/m; LAF-1 in buffer gave 34 Pa s. Frontier Two orders of magnitude separate condensates that are described in the literature with the same word, and many condensates take more than 100 s to fuse while showing mobile fractions below 50%, which is not liquid behaviour in any ordinary sense.

Established The best-characterised natural condensate turned out to be two phases, not one. In C. elegans P granules, PGL-3 behaves as a dynamic liquid with a fluorescence-recovery rate of 0.046 ± 0.008 s−1 and an exchange half-life near 22 s, while MEG-3 behaves as a gel: recovery 0.0070 ± 0.0007 s−1, half-life about 128 s, no detectable recovery in vitro, persistence of about 64% of the signal on dilution into buffer, resistance to 1 M NaCl, ATP and 1,6-hexanediol, and dissolution only in SDS. Frontier The gel scaffolds the liquid; the compartment that founded the field is a composite object, and the single-phase droplet language never described it.

Frontier In cells, the strongest negative result concerns heterochromatin, and it is specific rather than rhetorical. A 2020 study of mouse chromocentres reported that the foci “lack hallmarks of liquid droplets and rather resemble collapsed polymer globules”, that their size, accessibility and compaction are independent of HP1, and that they adopt two discrete folding states. Frontier That is a direct challenge to one of the three 2017 results that made phase separation a general theory of nuclear organisation, and it has not been withdrawn.

Frontier The most consequential in-cell disagreement is about whether the saturation concentration exists inside a cell at all. A January 2020 Science paper reported that droplet formation buffers the dilute-phase concentration and reduces expression noise, with noise rising more than twofold when droplets dissolved at mitosis and falling again when they reformed, in both an engineered DDX4 construct and endogenous NPM1. Frontier A May 2020 Nature paper measuring dense and dilute phases of nucleoli, Cajal bodies, stress granules and P-bodies concluded the opposite for endogenous compartments: heterotypic multicomponent interactions dominate, and these condensates “do not exhibit a fixed saturation concentration”. Speculative Both results are careful, both come from laboratories at the centre of the field, and they are four months apart; the field has never resolved them, and every buffering argument downstream inherits the ambiguity.

Established The field has formally acknowledged its methods problem, in writing, with names attached. On 19 August 2025 more than thirty authors led by Simon Alberti and Tanja Mittag published a community comment in Nature Communications stating that 1,6-hexanediol “should thus be avoided and not be used as evidence”, that FRAP readouts “alone are not directly informative regarding material states”, and that liquid–liquid phase separation and oil-in-water analogies “can be misleading”. Established It asks for proteins to be studied at endogenous expression in the relevant cell type, for in vivo phase diagrams over a wide concentration range, and for condensate complementation as the standard of functional proof. Frontier A field that publishes a document telling its own members to stop using a reagent is a field in better health than one that does not; it is also a field admitting that a large fraction of its published evidence used that reagent.

Frontier The reagent is still in use. An eLife reviewed preprint of March 2025 on oestrogen-responsive transcription at the TFF1 locus disrupted phase separation with 1,6-hexanediol and used the resulting change in transcript levels as part of its argument; the reviewers rated the evidence solid while noting the absence of direct mechanistic proof of condensation. Speculative Five months separate that paper from the consensus document that retired its main perturbation, which is the honest measure of how fast a methodological correction propagates.

Established Institutionally the subject is established beyond argument. The 2023 Breakthrough Prize in Life Sciences, announced 22 September 2022, went to Anthony Hyman and Clifford Brangwynne, $3 million each, for “a fundamental mechanism of cellular organization mediated by phase separation of proteins and RNA into membraneless liquid droplets”. Frontier The prize citation asserts the mechanism; the community comment published three years later asks for the evidence standards that would test it. Both statements are true, and the gap between them is this brief.

4 · Frontier questions

Frontier Question one, and the one everything else waits on: is phase separation necessary for any cellular function, or is it a consequence of the interactions that are? Multivalent binding concentrates molecules whether or not a phase boundary forms; a cluster of transcription factors and a demixed droplet look similar down a microscope and are different objects thermodynamically. Speculative The null hypothesis — that condensation accompanies function without causing it — has never been excluded for most of the compartments the field claims.

Frontier Question two: does the saturation concentration exist in a cell? The 2020 buffering result and the 2020 composition-dependence result disagree, and the disagreement is not about data quality. Speculative If endogenous condensates are multicomponent and heterotypic, then the single control parameter the whole framework is built around — one protein crossing one threshold — is a laboratory simplification and not a cellular mechanism. Handwave Nobody has proposed an experiment that would settle this for a named endogenous compartment, and until someone does, “above csat” is a phrase with two meanings in the same literature.

Frontier Question three: are transcriptional condensates real, and if they are, do they help? A February 2024 review in Communications Biology collects the contradiction: transcription-factor condensates colocalise with Med1, BRD4, H3K27ac and RNA polymerase II at target genes, and yet one study found that only 4% of transcription-factor clusters colocalise with nascent RNA signal, with effects ranging from inhibition to sustained activation, while a yeast experiment found that activation of a reporter did not depend on clustered recruitment at all. Speculative A mechanism that is present at 4% of the structures named after it, and whose removal sometimes increases output, is not yet a mechanism.

Frontier Question four: do material properties carry function, or only correlate with it? This is the one area where a causal experiment exists. An April 2024 Nature Communications study used light-inducible constructs to recruit specific messenger RNAs into condensates whose stiffness could be tuned by composition: 83% of target beta-actin transcripts were recruited, translation output fell about 39% over 12 hours, and solidifying the condensate strengthened the repression relative to a liquid one. Established In neurons, light-induced condensation reduced the spine-volume increase during long-term potentiation by 42%. Frontier The authors report the repression was weaker than the sequestration predicted, because individual messenger-RNA interactions with the condensate lasted under a second — a result that argues against the simple sequestration picture even as it supports a material-property effect.

Speculative Question five: is the two-phase vocabulary the right one? The 2025 community comment proposes coupled associative and segregative phase transitions as the organising concept and asks the field to stop saying liquid–liquid phase separation by default. Frontier The nanocluster result of November 2025 adds a state between monomer and droplet with condensate-like exchange kinetics. Handwave If there are three states rather than two, most published phase diagrams describe a boundary that is not where the biology happens.

Speculative Question six: is condensation an evolved trait or a physical inevitability? A 2025 Annual Review of Cell and Developmental Biology survey argues that condensation principles are used across organisms from bacteria to mammals and may have served as primitive compartments in prebiotic chemistry, and notes that fully structured proteins, not only disordered ones, can drive condensation. Handwave Ubiquity is equally consistent with selection and with the fact that concentrated polymer solutions demix whether or not anything wants them to, and the review does not separate the two. The prebiotic half of that argument belongs to Prebiotic Chemical Networks.

Speculative Question seven: can a condensate be drugged as a condensate? Dewpoint’s β-catenin programme is the test case, and its stated mechanism — preferentially forcing β-catenin into an inactive condensate in cancer cells — is a claim about partitioning rather than about occupancy. Handwave Nothing in a Phase 1a/2a safety and tolerability readout distinguishes a condensate mechanism from an ordinary allosteric one, so a clinical success would not by itself be evidence for this brief’s subject.

5 · Technological bottlenecks

Frontier The binding bottleneck is that there is no rheology inside an intact cell. Viscosity and surface tension are the quantities that make a condensate a phase rather than a cluster, and the methods that measure them — micropipette aspiration, optical-tweezer fusion, active microrheology — work on droplets in a dish. Established The 2025 community comment lists what is available in cells: single-particle tracking, fluorescence correlation spectroscopy, Brillouin microscopy, bending-rigidity analysis and contact-angle measurement at membranes. Speculative None of those returns a number in pascal-seconds with the confidence that the in vitro methods do, which is why the comparison table in this subject runs from about 1 to about 34 Pa s and nobody treats the spread as an error bar.

Frontier The second is that the standard perturbations are non-specific, and the field has said so. 1,6-hexanediol dissolves condensates and also has, in the consensus document’s words, “various detrimental effects in cells”. Established Fluorescence recovery after photobleaching measures exchange, which depends on binding, geometry and photophysics as much as on material state. Handwave Round shape, fusion and fast recovery were the three pillars of a decade of in-cell claims, and all three are now formally disclaimed by the people who built them.

Frontier The third is the tag. A saturation concentration of 0.5 to 1 µM for pre-cleaved FUS and 2 to 5 µM for tag-cleaved FUS is a factor of five introduced by a purification protocol, and both numbers are in the range that a cell biologist would call physiological. Speculative Any argument of the form “the cellular concentration exceeds csat, therefore the condensate forms” is sensitive to a choice made at the bench, and the literature rarely states which choice was made.

Frontier The fourth is the concentration regime. Reconstitutions that made a protein canonical were sometimes run one to two orders of magnitude above any plausible cellular concentration — 100 µM for the DDX4 disordered region, against 800 nM for LAF-1 — and crowding agents can induce demixing non-specifically. Speculative A 2026 review states plainly that high concentrations of inert polymeric crowders may non-specifically induce phase separation, which means a positive in vitro result at high crowder is weak evidence about a cell.

Frontier The fifth is optical. The consensus recommendations split imaging by size: wide-field or confocal above 300 nm, super-resolution between 20 and 300 nm. Speculative The FUS nanoclusters at 300 to 500 nm sit at that boundary, and most published in-cell condensate work used the wide-field regime. Handwave A field whose central object may be smaller than its dominant imaging modality has a systematic blind spot rather than a noise problem.

Speculative The sixth is that overexpression is the default and the comment says it should not be. A 2026 review notes the field’s heavy reliance on overexpression systems and says this leaves open whether condensates form at endogenous expression levels at all. Handwave That is not a caveat about precision. It is a caveat about whether the phenomenon is there.

Frontier The seventh is commercial and it has already bitten. Faze Medicines raised $81 million, ran for twenty-three months, never entered the clinic and closed in November 2022; its investor said the science did not progress sufficiently. Speculative The bottleneck a drug programme hits is not whether condensates exist but whether a molecule can change one selectively, and a failed company is the only public data point on that question until DPTX3186 reads out.

6 · Research dependencies

Established This subject depends on instrumentation and on statistics, not on results from other briefs. The measurements that matter come from optical tweezers, micropipette aspiration, super-resolution microscopy, single-particle tracking, cryo-electron tomography and quantitative fluorescence calibration — techniques developed for soft-matter physics and structural biology and applied here. Frontier Nothing on this map blocks this brief.

Frontier The dependency with the clearest near-term payoff is in-situ structural biology. A December 2024 preprint used high-pressure freezing, focused-ion-beam milling and cryo-electron tomography to resolve nucleosomes inside reconstituted chromatin condensates at 6.1 angstroms, and at 12 angstroms in native chromatin, finding a nearly random nucleosome orientation distribution in both. Speculative Structural randomness inside a dense compartment is what a liquid should look like, and this is the first time anyone could check at that resolution; one preprint is not a result, and the reconstituted case is not a cell.

Speculative The dependency nothing supplies is a way to change phase behaviour without changing interaction chemistry. Every existing tool — charge scrambling, aromatic-residue substitution, disordered-region truncation, tag fusion, optogenetic oligomerisation — moves valency or affinity as a side effect. Handwave Until a mutation exists that moves only the demixing threshold, the field’s central claim is not directly testable, and every functional assignment is an inference from a perturbation that did more than one thing.

Frontier Traffic with the rest of this map runs outward. Neurodegeneration Reversal inherits the liquid-to-solid transition as a candidate mechanism for FUS and TDP-43 pathology; Theories of Carcinogenesis inherits the claim that oncogenic transcription factors work through condensates; RNA Medicines and Synthetic Biology inherit condensation as an engineering primitive. Speculative Each of those inheritances is only as good as the causality argument this page says has not been made.

7 · Required experiments

Frontier The tests below rank by what a result would settle, and the field’s own consensus document names the top one.

Speculative Tier one, and the decisive experiment for this brief: a separation-of-function mutant that abolishes phase behaviour at unchanged binding affinity and valency, in a cell expressing the protein at endogenous level, with a measured function that fails only then. Frontier The requirement is exact: the mutant must have the same partners, the same dissociation constants and the same number of interaction sites as the wild type, and must differ only in where its demixing threshold sits. Established The nearest existing approach is the charge-scrambled DDX4 disordered region, which holds amino-acid composition constant and abolishes organelle formation, but changes the interactions in doing so. Handwave No published mutation satisfies the strict version of this condition for any endogenous condensate, which is why a framework with a Breakthrough Prize attached still has a contested causal claim at its centre.

Established Tier one has a complementation variant that the field has formally endorsed, and it is easier to build. The August 2025 community comment asks for loss-of-function disruption of phase separation followed by rescue with an orthogonally encoded phase-separation activity, and states that restoration of the wild-type phenotype by that route is “strong evidence for a functional role of condensation”. Frontier The logic is sound and the control is the hard part: the orthogonal module must reconstitute the phase behaviour without reconstituting the specific chemistry that was removed.

Frontier Tier two: an in vivo phase diagram for one named endogenous compartment, at endogenous expression, across a wide concentration range. This is the experiment that would settle whether a saturation concentration is a cellular quantity, and it is the direct empirical arbiter between the 2020 buffering result and the 2020 composition-dependence result. Speculative It requires absolute concentration calibration in living cells, controlled titration of a single component without perturbing others, and quantitative imaging of both phases. Speculative Every element of that exists; the combination for one compartment, published as a phase diagram with error bars, does not.

Frontier Tier three: in-cell rheology that returns a number. Brillouin microscopy, active microrheology with optically trapped probes, and bending-rigidity analysis of condensate interfaces are the candidates the community comment lists. Speculative A viscosity for a named condensate in an unperturbed cell, with an uncertainty, would do more for this subject than another ten reconstitutions, because it is the measurement that would let the in vitro record be checked rather than merely extended.

Established Tier four is running and is the template for how this should be done: tune the material state, hold the composition, read out a function. The 2024 optogenetic messenger-RNA experiment recruited 83% of target transcripts, measured a 39% fall in translation over 12 hours, showed that gel-like condensates repressed more strongly than liquid ones at matched recruitment, and reported a 42% reduction in spine-volume growth during potentiation in neurons. Frontier Its own weakness is instructive: individual messenger-RNA residence times under one second mean the functional effect is not simple sequestration, and the authors say so.

Speculative Tier five: measure the nanocluster state in cells and find out whether the dilute phase is dilute. The November 2025 result puts 300 to 500 nm clusters at 200 nM in vitro, below the saturation concentration, with exchange above 3,000 molecules per square micron per second. Frontier The in-cell version needs super-resolution in the 20 to 300 nm band at endogenous expression. Handwave If clusters at sub-saturation carry the biochemistry, then the droplets everyone has been imaging are a late and possibly incidental stage.

Frontier Tier six: settle the heterochromatin case, because it is the one place where a clean negative was published and not retracted. The 2020 chromocentre study found compaction, size and accessibility independent of HP1 and two discrete folding states rather than droplet behaviour. Speculative A pre-registered replication contest on a single locus, with both camps agreeing the readouts in advance, would be cheap and would be the most informative thing the field could do with a year.

Speculative Tier seven is clinical and will happen whatever the biology says. DPTX3186 entered a multi-centre dose-escalation Phase 1a/2a in advanced solid tumours with a gastric-cancer focus, first patient dosed 4 February 2026, carrying FDA orphan-drug and fast-track designations, with translational biomarkers for pathway engagement. Handwave A biomarker showing β-catenin redistribution would be suggestive; only a comparison against a matched molecule that inhibits the same target without altering partitioning would make the trial evidence about condensates.

Established And the experiments that are reported as tests of this subject and are not. Dissolving a structure with 1,6-hexanediol, observing that it is round, watching two of them fuse, and showing fast fluorescence recovery are, by the field’s own August 2025 statement, not evidence of phase separation. Speculative They remain in the literature in large numbers, including in papers published months after that statement, and any reader assessing a condensate claim should check which of these four it rests on.

8 · Engineering requirements

Frontier There is one engineering programme downstream of this page and it is pharmaceutical. Everything else in this section is instrumentation for answering a question rather than building a product.

Speculative The drug specification, as its sponsor states it. DPTX3186 is a first-in-class small molecule intended to force β-catenin into an inactive condensate preferentially in cancer cells, with the stated aim of avoiding the bone and gastrointestinal toxicity that has ended previous Wnt-pathway programmes; the sponsor projected clinical entry by the end of 2025, early proof of concept by the end of 2026, and runway into early 2027. Frontier A second oral candidate disrupting MYC condensates was reported at a cancer-research meeting in April 2026 with tumour regression and stasis in solid-tumour models and selective killing of high-MYC cells. Handwave Both descriptions are company statements without published dose-response data, and neither has been shown to act through a condensate rather than through the target.

Established The measurement specification is the part with numbers. An instrument adequate to this subject needs: absolute concentration calibration in live cells; imaging resolution below 300 nm and ideally into the 20 to 300 nm band; single-particle tracking for intra-condensate diffusion; a rheological readout with an uncertainty; composition mapping by crosslinking or proximity labelling with mass spectrometry and matched controls; and induction that is reversible on a timescale shorter than the process being studied. Frontier Every item on that list exists separately and the combination is what the 2025 community comment is asking for.

Speculative Engineered condensates are the reverse direction and are further along than the biology. CD-CODE already catalogued 375 synthetic in vitro condensates in 2023, and optogenetic and chemically induced systems can create a compartment on demand in a chosen location. Frontier The community comment’s own caveat is the engineering requirement: an induced condensate must be shown to match the endogenous one in location, size, composition and material properties before it can stand in for it. Handwave Most published induced systems do not report all four. This is the seam with Synthetic Biology and with Biological Computing.

9 · Adjacent technologies

Established The adjacent fields supplying the evidence on this page are soft-matter physics and microscopy, not cell biology. Polymer theory supplies the sticker-and-spacer framework and the Flory–Huggins parameter that organises every phase diagram in the subject; micropipette aspiration and optical tweezers supply the material properties; cryo-electron tomography supplies the only structural data. Frontier Cell biology supplies the claims.

Frontier Within this map the closest page is Embryo Models, and the two are deliberately inverse. That brief is about self-organisation at tissue scale, where the structures are visible, the fidelity question is about how closely a model matches a real embryo, and the binding constraint is regulatory. This page is about an organising principle inside a single cell, where the structures are at or below the diffraction limit and the binding constraint is that nobody can perturb the principle without perturbing everything else. Speculative The shared question is the same one: when a structure forms by itself, how do you show it matters.

Speculative Three neighbours inherit something concrete and one inherits a warning. Neurodegeneration Reversal inherits the liquid-to-solid transition as the leading physical account of how a soluble RNA-binding protein becomes a fibril. Theories of Carcinogenesis inherits the transcriptional-condensate account of oncogene addiction and now a clinical asset to test it with. RNA Medicines inherits the finding that RNA changes condensate viscosity by threefold at micromolar concentrations, which is a delivery-relevant fact about what happens when exogenous RNA arrives in a cell. Handwave Generative Biomolecular Design inherits the warning: models that predict binding do not predict demixing, and a designed sequence that forms a condensate in silico has not been shown to do anything in a cell.

Speculative One adjacency runs the other way and is the most direct. Noncoding Genome Function asks what fraction of the genome does anything, and condensation is one of the few routes by which an unconserved transcript could do real work without leaving a selection signature: RNA changes LAF-1 droplet viscosity threefold at micromolar concentration, and a scaffolding role need not constrain sequence. Handwave That is a mechanism for a claim rather than evidence for it, and neither page should be read as supplying the other’s answer.

Frontier One adjacency is methodological and it is the most useful one. Anaesthetic Mechanism and Sleep Function share this subject’s structure exactly: a phenomenon everyone agrees is real, a mechanism proposed from physics, and a causality claim that resists the obvious experiment because the perturbation always does more than one thing. That is the register of Category XI, and it is why these pages sit together.

10 · Institutional requirements

Established This field’s institutional problem is that it became established before its evidence standards did, and it has now written that down. The 2023 Breakthrough Prize, the 244-condensate database, the dedicated conference circuit and the venture capital all predate the August 2025 document that retired the field’s most common perturbation. Frontier That ordering is the institutional fact that matters, and it is not unique to this subject.

Speculative The cheapest useful institutional intervention is a reporting standard, and it does not exist. A condensate claim should state: the expression level relative to endogenous, the tag and whether it was cleaved, the concentration regime, the crowder if any, the imaging modality and its resolution, the perturbation and its known off-target effects, and whether an in vivo phase diagram was attempted. Frontier Every item on that list is in the 2025 comment as a recommendation. Handwave A recommendation without a journal checklist changes practice slowly, and the five-month gap between the comment and papers still using hexanediol is the measurement of how slowly.

Frontier The funding structure rewards the wrong experiment. Demonstrating that a new protein forms condensates is publishable, quick and additive; demonstrating that an existing condensate claim fails a separation-of-function test is slow, adversarial and hard to place. Speculative The 244 entries in CD-CODE are almost entirely positive findings, because that is what the incentive structure produces, not because negatives are rare.

Established The commercial institution has already demonstrated both failure modes. Faze Medicines raised $81 million from Third Rock, Novartis, Eli Lilly, AbbVie, Invus, Catalio, Casdin and Alexandria, launched in December 2020, and closed in November 2022 without a clinical candidate. Established Dewpoint raised about $287 million by early 2023, partnered with Bayer, Merck, Pfizer and Volastra, reported more than twenty programmes, and reached first-in-human dosing three years after its stated target. Frontier Nereid Therapeutics and Transition Bio each raised $50 million Series A rounds in the same window. Speculative About half a billion dollars of private capital has been committed to a causal claim that the field’s own consensus document says is usually not tested properly, and that is the institutional risk this page exists to name.

11 · Ethical & societal considerations

Frontier The ethical issues here are about evidence rather than about subjects. There are no human participants in a phase diagram and no dual-use pathway in a droplet, so the obligations that bite are the ones about what a field owes when it has advertised a mechanism ahead of the demonstration.

Speculative The first is a patient-facing one and it is live now. A condensate-modulating molecule in a Phase 1a/2a oncology trial recruits patients on the strength of a mechanistic story. Frontier Advanced gastric cancer has poor options and a novel mechanism is a reasonable thing to try; the obligation is that the consent conversation describes the asset as a small molecule affecting β-catenin, which is measurable, rather than as a condensate therapy, which is an interpretation. Handwave There is no evidence that any sponsor has done otherwise, and the point is prospective.

Frontier The second is about correction. A field that publishes a consensus document retiring a reagent has an obligation to say what that means for the results obtained with it, and the document does not do that. Speculative Somewhere in the literature is a large set of conclusions that rest on hexanediol and on roundness, and nobody has counted them; a systematic audit would be unwelcome, cheap and more valuable than most new experiments.

Speculative The third is about students. A doctoral project built on demonstrating that protein X forms condensates is a project whose central claim may be retired by a methods standard published during the thesis. Handwave Supervisors who direct trainees into the additive version of this field rather than the testing version are making a career allocation on their behalf, and it is a worse bet in 2026 than it was in 2019.

Frontier The fourth is about the public account. Prize citations and press releases state the mechanism as fact; the consensus document states the evidence standards as aspirations. Speculative Both documents are honest, and a reader outside the field has no way to know that they describe different epistemic situations, which is a communication failure rather than a scientific one.

12 · Civilizational implications

Speculative If condensation is causal, the consequence is a second layer of cellular regulation that is thermodynamic rather than informational, and the honest statement about what follows is that nobody knows. Genetics, signalling and metabolism are described in terms of which molecule binds which; a phase layer would be described in terms of which components are together, at what concentration, in what material state. Frontier Those are different kinds of explanation and the textbooks have room for only one of them at present.

Speculative The one concrete consequence is a class of drug targets that occupancy pharmacology has not reached. Transcription factors, including β-catenin and MYC, have resisted small-molecule inhibition for three decades because they work through extended, low-affinity, disordered interfaces. Frontier Altering where such a protein partitions is a different intervention from blocking a pocket it does not have. Handwave Whether that difference is real is what one Phase 1a/2a trial and one preclinical MYC programme will begin to indicate, and neither will settle it.

Speculative A second consequence is about the origin of life and it runs backwards through the argument. If demixing is what concentrated prebiotic chemistry before membranes, then compartmentalisation preceded heredity and the first compartments cost nothing to build. Handwave That story is coherent, has been argued in a 2025 review, and has no experiment attached that distinguishes it from the alternatives; it belongs to Prebiotic Chemical Networks and is carried here only as an inheritance.

Frontier And the outcome that is more likely than either: the framework is partly right, and the correction is a narrowing rather than a collapse. A plausible end state is that a small number of compartments — the nucleolus, stress granules, germ granules — are established as phase-separated with causal consequences, while transcription, heterochromatin and signalling are re-described as multivalent clustering that does not cross a phase boundary. Speculative That would leave the physics intact, the vocabulary narrowed, and roughly a decade of generalisation to be quietly unwritten, which is the ordinary way a framework corrects and the least discussed possibility in the field.

13 · Timelines

These horizons track evidence standards and one clinical readout, because there is no engineering programme here to track:

  • 10 yr: Frontier Expect the 2025 community recommendations to become journal practice, expect hexanediol to disappear from methods sections, and expect a first in vivo phase diagram for a named endogenous compartment at endogenous expression. Speculative Expect the β-catenin and MYC condensate programmes to read out, and expect the result to be interpretable as ordinary pharmacology whichever way it goes. Speculative Expect the sub-saturation nanocluster state to be either confirmed in cells or shown to be an in vitro artefact, and expect that to matter more than any clinical result on this horizon.
  • 25 yr: Speculative This is the window in which the decisive experiment plausibly runs: a separation-of-function allele at endogenous expression with a function that fails only when the phase behaviour does. Speculative A positive result would make condensation a regulatory layer alongside transcription and phosphorylation. Speculative A null for the compartments where it is tried would narrow the framework to the ribonucleoprotein granules where it began, which is the modal outcome.
  • 50 yr: Handwave On this horizon the plausible development is that the phase question stops being asked separately, because a quantitative description of a cell that includes local concentration and local material state makes “is it a phase?” a question about a parameter value rather than about a category. Speculative Nanomedicine and Precision Medicine would inherit that description if it arrives.
  • 100 / 250+ yr: Handwave Beyond useful forecasting. The defensible statement is about shape rather than date: an idea that explains everything in its domain within fifteen years of being proposed either acquires a decisive test or becomes a vocabulary, and this one has had the test specified and not performed. Handwave Any claim about what a mature condensate engineering discipline would build is an assertion, and this page does not make one.

14 · Technology tree & dependencies

  • Depends on No typed dependency edge to another brief on this map, and the absence is informative. This subject waits on instrumentation and on a perturbation nobody has built, neither of which any page here produces. Embryo Models is the nearest page and supplies a parallel rather than a prerequisite: self-organisation at tissue scale, with the same problem of proving that a structure which forms by itself is doing the work. Generative Biomolecular Design builds the sequence models that would have to predict a demixing threshold, and is not blocking, because those models are being built for binding.
  • Requires (not on this map) Six conditions, in the order of the tokens above. The mutation. An allele with the same partners, dissociation constants and valency as the wild type that moves only the demixing threshold; every existing tool — charge scrambling, aromatic substitution, truncation, tag fusion, optogenetic oligomerisation — changes affinity or valency as a side effect. The phase diagram. One named endogenous compartment, titrated across a wide concentration range at endogenous expression with absolute calibration, which is the only thing that would arbitrate between the 2020 buffering result and the 2020 composition-dependence result. The rheometer. A viscosity in pascal-seconds for a condensate in an unperturbed cell, with an uncertainty; the in vitro spread runs from about 1 to about 34 Pa s and nobody can check it in situ. The microscope. Imaging at 20 to 300 nm as routine practice rather than as a specialist capability, because the FUS nanocluster state at 300 to 500 nm sits below most published in-cell work. The standard. A checklist requiring expression level, tag state, concentration regime, crowder, imaging resolution and perturbation off-targets to be declared — the cheapest item on this list and the one whose absence does the most damage. The clinical control. A matched molecule inhibiting the same target without altering partitioning, without which no trial outcome is evidence about condensates.
  • Enables A positive separation-of-function result would give Theories of Carcinogenesis a mechanism for oncogene addiction that is currently an analogy, and would give Neurodegeneration Reversal a physical account of fibril nucleation with a tunable parameter. A null would narrow the framework rather than eliminate it, and no typed enabling edge is claimed, because a causal claim that has not been demonstrated enables nothing on a schedule.
  • Adjacent Soft-matter physics, super-resolution microscopy, cryo-electron tomography and quantitative fluorescence calibration supply every number on this page. Within this map: Embryo Models for self-organisation at the next scale up, RNA Medicines for what exogenous RNA does to condensate material properties, Synthetic Biology and Biological Computing for engineered compartments, Prebiotic Chemical Networks for the origin-of-life inheritance, Noncoding Genome Function for whether an unconserved transcript can do work by scaffolding a compartment, and Anaesthetic Mechanism and Sleep Function for the shared epistemic shape.

15 · Common misconceptions & speculative claims

Established “Round, fusing, and fast-recovering means phase-separated.” It does not, and the field said so on 19 August 2025. Surface tension makes many things round; apparent fusion can be two structures moving along a shared template; fluorescence recovery depends on binding kinetics, geometry and photophysics. Frontier The consensus document states that FRAP readouts alone are not directly informative regarding material states, which retires the single most common figure panel in the field’s first decade.

Established “1,6-hexanediol dissolves condensates, so if it dissolves a structure that structure is a condensate.” The 2025 comment states that hexanediol has various detrimental effects in cells and “should thus be avoided and not be used as evidence”. Speculative The reagent appears in papers published after that statement, so this misconception is not historical.

Frontier “Condensates are liquid droplets.” Some are; the founding compartment is not. P granules contain a gel-like MEG-3 scaffold with an exchange half-life of about 128 seconds around a liquid PGL-3 phase with a half-life of about 22 seconds. Established Many condensates take more than 100 seconds to fuse and have mobile fractions under 50%. Frontier The consensus document asks the field to stop defaulting to liquid–liquid phase separation as a description, which is an unusual thing for a field to say about its own name.

Frontier “Phase separation explains transcription.” It may contribute. The measured record includes a study in which only 4% of transcription-factor clusters colocalised with nascent RNA, effects ranging from inhibition to activation, and a yeast experiment in which reporter activation did not depend on clustered recruitment. Speculative The honest statement is that transcriptional condensates are a live hypothesis with disconfirming data attached, not an established layer of gene regulation.

Frontier “Heterochromatin is a phase-separated compartment.” One well-cited 2020 study of mouse chromocentres reported the opposite: no hallmarks of liquid droplets, behaviour resembling collapsed polymer globules, and size, accessibility and compaction independent of HP1. Speculative The claim and its rebuttal are both in the literature and the field has not adjudicated between them; a reader who has only met one of them has met half the evidence.

Speculative “Cellular concentrations exceed the saturation concentration, so condensates form.” The saturation concentration for FUS differs by about fivefold depending on whether the solubility tag was cleaved, and a November 2025 study reports that the unstressed cellular condition is commonly sub-saturated, with nanoclusters rather than droplets. Handwave An inequality between two numbers of uncertain provenance is not a mechanism.

Speculative “Condensate drugs are a new therapeutic modality.” They are, at present, small molecules with a mechanistic hypothesis attached. Frontier One is in a Phase 1a/2a trial and one is preclinical; a company with $81 million and the same thesis closed in 2022 without a candidate. Handwave Until a trial includes a matched comparator that hits the same target without altering partitioning, “condensate modulator” is a description of intent rather than of demonstrated mechanism.

Frontier “Phase separation is a discredited fad.” This is the opposite error and it is also wrong. Established Purified LAF-1 demixes above about 800 nM into droplets of 34 Pa s; RGG condensates have a surface tension of 0.159 mN/m; DDX4 droplets concentrate single-stranded DNA and exclude double-stranded DNA. Established The physics is measured and reproducible. What is contested is the inference from the physics to the biology, and collapsing that distinction in either direction is the characteristic mistake about this subject.
